PEGs for mRNA Delivery

Messenger RNA (mRNA) has emerged as a new class of therapeutic agent for the prevention and treatment of various diseases. In order to function in vivo, mRNA requires a safe, effective and stable delivery system that protects the nucleic acid from degradation and allows for cellular uptake and release of mRNA. Lipid nanoparticles (LNPs) have successfully entered the clinic for delivery of mRNA; in particular, lipid nanoparticle-mRNA vaccines are now approved against COVID-19, marking a milestone in mRNA therapy.

Lipid nanoparticles(LNPs) are typically composed of four components. (1) cationic or ionizable lipids, which are smart macromolecules that can be functionalized to improve mRNA entrapment, enhance cellular uptake efficiency, and promote endosomal escape; (2) lipid-anchored polyethylene glycol (PEG), which is used to reduce particle size, prevent particle aggregation, increase circulation time, and reduce uptake by non-target cells; (3) cholesterol that stabilizes the LNPs complex; and (4 ) phospholipids that support the lipid bilayer structure.

Huateng Pharma  is able to provide the following PEG products for your mRNA delivery from mg to kg. PEG linkers with molecular weights and functional groups not listed in our online catalog may be available by custom synthesis.
